ABSTRACT

The military vocational education is facing the reform under the background of "Internet +" education. O2O teaching is a model that combines the online teaching resource platform with the offline traditional education, and it is consistent with the development trend of contemporary military vocational education. In this study, we analyze the comparative advantages of O2O teaching model in military vocational education, including the multi-dimensional teaching content, the interactive teaching method, and the autonomy of teaching design. In this model, students become the subject of learning, and the process of storing knowledge will be transformed into the application and creation of knowledge. This paper also elucidates the feasibility of O2O teaching in the military vocational education, and further discusses its design in application from the point of view of managers, teachers and learners.

ABSTRACT

Cinnamomum camphora is an important economic tree species in China. According to the type and content of main components in the volatile oil of leaf, C. camphora were divided into five chemotypes, including borneol-type, camphor-type, linalool-type, cineole-type, and nerolidol-type. Terpene synthase(TPS) is the key enzyme for the formation of these compounds. Although several key enzyme genes have been identified, the biosynthetic pathway of(+)-borneol, which has the most economic value, has not been reported. In this study, nine terpenoid synthase genes CcTPS1-CcTPS9 were cloned through transcriptome analysis of four chemical-type leaves. After the recombinant protein was induced by Escherichia coli, geranyl pyrophosphate(GPP) and farnesyl pyrophosphate(FPP) were used as substrates for enzymatic reaction, respectively. Both CcTPS1 and CcTPS9 could catalyze GPP to produce bornyl pyrophosphate, which could be hydrolyzed by phosphohydrolase to obtain(+)-borneol, and the product of(+)-borneol accounted for 0.4% and 89.3%, respectively. Both CcTPS3 and CcTPS6 could catalyze GPP to generate a single product linalool, and CcTPS6 could also react with FPP to generate nerolidol. CcTPS8 reacted with GPP to produce 1,8-cineol(30.71%). Nine terpene synthases produced 9 monoterpene and 6 sesquiterpenes. The study has identified the key enzyme genes responsible for borneol biosynthesis in C. camphora for the first time, laying a foundation for further elucidating the molecular mechanism of chemical type formation and cultivating new varieties of borneol with high yield by using bioengineering technology.

ABSTRACT

OBJECTIVE To evaluate the intervention effect of Caulis sinomenii compatible with prepared Aconiti Lateralis on bone destruction in rheumatoid arthritis (RA)model rats ,and to investigate its mechanism. METHODS Totally 40 SD rats were randomly divided into blank group ,model group ,positive control group (indomethacin 0.013 5 g/kg)and C. sinomenii compatible with prepared Aconiti Lateralis group (C. sinomenii 1.08 g/kg+prepared Aconiti Lateralis 1.35 g/kg)according to body mass ,with 10 rats in each group. Except for the blank group ,all the other groups made RA rat models by injecting type Ⅱ bovine collagen. Rats in each group were given corresponding drugs or distilled water intragastrically. The general information ,body weight ,foot swelling and arthritis index (AI)scores of rats in each group were recorded. After the 30th day of administration ,the changes of ankle bone in rats were detected by small animal CT machine. The levels of inflammatory factors [interleukin- 31(IL-31),IL-25 and IL- 3] and chemokines [receptor activator of nuclear factor κB ligand(RANKL),receptor activator of nuclear factor κB (RANK)and osteoprotegerin (OPG)] in serum were detected by enzyme-linked immunosorbent assay. Pathological indexes of rat ankle joint were observed by HE staining. Immunohistochemical method was used to detect the expression of RANKL ,RANK and OPG in synovial tissue of rat ankle joint. RESULTS Compared with blank group ,the mental state of the model group was weak , the activity decreased significantly ,the hair lost luster ,and the body weight decreased significantly on the 12th to 30th days (P< 0.05 or P<0.01);the swelling degree of the foot was significantly increased and the AI score was significantly increased on the 12th to 30th days(P<0.01);the ankle joint in model group had rough surface ,obvious tissue damage and serious bone erosion ; serum levels of IL- 31,IL-25,IL-3,RANKL and RANK were increased significantly ,while the level of OPG was decreased significantly (P<0.01); the expression of RANKL and RANK in synovium of ankle joint increased significantly , while the expression of OPG decreased significantly (P<0.01). Compared with model group ,the above indexes of administration groups were improved to varying degrees ,and most of the differences were statistically significant (P<0.05 or P<0.01). CONCLUSIONS By inhibiting the RANKL/RANK/OPG signaling pathway ,C. sinomenii compatible with prepared Aconiti Lateralis can inhibit the excessive proliferation of osteoclasts and restore the balance of bone metabolism so as to play a role in protecting bone joints and treating RA.

ABSTRACT

At present, the disjointing situation is generally present between "systemic"and "local", "macro" and "micro", " process" and "activity evaluation" in the study of traditional Chinese medicine (TCM). An urgent task for the modernization of TCM is to establish new strategies and methods which can reflect the overall characteristics of TCM. The introduction of integrative pharmacology provided a feasible approach to solve the problem of the fragmentation of TCM. Internet-based computation platform method was adopted in this study to explore the active molecular mechanism of Mylabris in the treatment of colorectal cancer. Based on the analysis of the functional integration of internet-based computation platform V1.0 version software, the "core components-key target- main pathway" multidimensional network of Mylabris in treatment of colorectal cancer disease was constructed to explore the potential molecular mechanism of Mylabris in treatment of colorectal cancer from multiple perspectives. The results showed that Mylabris can treat the colorectal cancer and the mechanism might be associated with amino acid metabolism, NF-κB signaling pathway, immune system, endocrine system, nervous system, and chemokine signal transduction pathway, epithelial cell signaling in helicobacter pylori infection, T cell receptor signaling pathway, B cell receptor signaling pathway and so on.

ABSTRACT

Objective To explore the underlying anatomy of iliac vein compression syndrome (IVCS) using CT, and discuss the imageological index for subtype diagnosis and potential clinical significance. Methods We retrospectively analyzed the imageological data of 69 IVCS patients from July, 2009 to June, 2014. According to CT findings, patients were categorized to simple IVCS (sIVCS, the iliac vein is compressed by only the anterior iliac artery, n=22), lumbar degeneration?related IVCS ( dIVCS, besides the iliac artery, the lower lumbar degenerative changes including osteophyte, protruded disc, etc. pressed the iliac vein from the back, n=33) and other IVCS causes (oIVCS, including tuberculosis, inflammation and fracture hematoma of the lumbar spine, n=14); meanwhile, 69 age? and sex?matched subjects was set as a control group. Evaluated indexes were onset age, course of lower limb swelling and pain, size of the iliac vein tunnel ahead lumbar (IVTAL), lower lumbar spine forward curvature angle (LLSCA), iliac vein?pressed signs by lower lumbar degeneration, compression sites, scope of deep venous thrombosis and interventional therapeutic effect. The differences of those indexes of various groups were compared. Data as onset age, course of disease, sizes of IVTAL and LLSCA were evaluated using variance analysis of the general linear model, with Bofferroni test correction for multiple comparisons. Data as iliac vein?pressed site, scope of venous embolism and therapeutic effect were assessed by crossing table χ2 test. All statistical analyses were performed using SPSS17.0 software (SPSS company, Chicago, USA). Bilateral P≤0.05 were considered to be significant. Results The onset mean age was (61.5 ± 10.6) yr. in dIVCS, (42.3 ± 6.5)yr in sIVCS. and(53.1 ± 16.8)yr. in oIVCS with a significant difference ( F=11.030, P<0.01). Mean sagittal diameter of the IVTAL and LLSCA were(2.3±0.5)mm and(121.8±5.4)° in dIVCS and(2.5± 0.5)mm and(124.4 ± 3.9)° in sIVCS, respectively; which were smaller than those of control group [(6.4 ± 1.6)mm and(127.5 ± 7.3)° , respectively ] and oIVCS [(5.9 ± 2.3)mm and(129.5 ± 5.9)° , respectively ](F=125.275,P<0.01 for sigittal diameter and F=7.95,P<0.01 for LLSCA). The degenerative changes compressing the iliac vein of dIVCS had 41 sites of 33 patients which were respectively the discal forward protrusion or bulge (51%, 17/33), vertebral anterior osteophyte (50%, 16/33)and lower lumbar vertebrae slippage (19%,8/33). The compression sites of dIVCS located exactly anterior to the fifth lumbar(18/33, 56%), the intervertebral disk between the fourth and fifth lumbar(9/33, 26%), the right front (3/33, 9%) and left front (3/33, 9%) of the fifth lumbar, however, that of sIVCS sited the right front(14/22, 64%), exactly the front(4/22, 18%)of the fifth lumbar and exactly anterior(4/22, 18%)to the intervertebral disk between the fourth and fifth lumbar, with a significant difference of location distribution(χ2=19.305, P<0.01). In sIVCSs, deep vein thrombosis of all exceeded the iliac vein length and implicated the femoral vein by 27%(6/22)and the popliteal vein by 73%(16/22), while in dIVCSs 18%(6/33)patients had deep vein thrombosis in the iliac vein, 30%(10/33)in the femoral vein and 52%(17/33)in the popliteal vein, and in oIVCSs, 30%(4/14)patients had deep vein thrombosis in the iliac vein, 40%(6/14)in the femoral vein and 30%(4/14)in the popliteal vein, with a significant difference of thrombosis scope(χ2=9.28, P<0.01). 86%sIVCSs needed intravenous stent?implanted operation to obtain effective treatment, only 52%dIVCSs were performed stent?implanted operation for effective therapy, none of oIVCSs had stent?implanted operation before the iliac vein recanalization. Conclusion CT can precisely display the pathological anatomy features of different IVCS patients, which can potentially help clinicians plan accurate treatment strategy.

ABSTRACT

OBJECTIVETo investigate the underlying metabolomic profifiling of coronary heart disease (CHD) with blood stasis syndrome (BSS).

METHODSCHD model was induced by a nameroid constrictor in Chinese miniature swine. Fifteen miniature swine were randomly divided into a model group (n=9) and a control group (n=6), respectively according to arandom number table. After 4 weeks, plasma hemorheology was detected by automatic hemorheological analyzer, indices including hematocrit, plasma viscosity, blood viscosity, rigidity index and erythrocyte sedimentation rate; cardiac function was assessed by echocardiograph to detect left ventricular end-systolic diameter (LVED), left ventricular end-diastolic diameter (LVEDd), ejection fraction (EF), fractional shortening (FS) and other indicators. Gas chromatography coupled with mass spectrometry (GC-MS) and bioinformatics were applied to analyze spectra of CHD plasma with BSS.

RESULTSThe results of hemorheology analysis showed signifificant changes in viscosity, with low shear whole blood viscosity being lower and plasma viscosity higher in the model group compared with the control group. Moreover, whole blood reduction viscosity at high shear rate and whole blood reduction viscosity at low shear rate increased signifificantly (P <0.05). The echocardiograph results demonstrated that cardiac EF and FS showed signifificant difference (P <0.05), with EF values being decreased to 50% or less. The GC-MS data showed that principal component analysis can clearly separate the animals with BSS from those in the control group. The enriched Kyoto Encyclopedia of Genes and Genomes biological pathways results suggested that the patterns involved were associated with dysfunction of energy metabolism including glucose and lipid disorders, especially in glycolysis/gluconeogenesis, galactose metabolism and adenosine-triphosphate-binding cassette transporters.

CONCLUSIONSGlucose metabolism and lipid metabolism disorders were the major contributors to the syndrome classifification of CHD with BSS.

ABSTRACT

BACKGROUND:Articular cartilage is a highly diferentiated tissue, which is very limited in its ability to repair after injury. Stem cel therapy for cartilage repair has completely solved this problem. OBJECTIVE: To investigate the mechanism that diferent growth factors induce the diferentiation of bone marrow mesenchymal stem cels into chondrocytes. METHODS:Passage 5 rat bone marrow mesenchymal stem cels were induced by diferent growth factors and their combinations, including transforming growth factor beta 1 (TGF-β1) group, TGF-β1+insulin growth factor-1 (IGF-1) group, and bone morphogenetic protein 2 (BMP-2)+IGF-1 group, TGF-β1+BMP-2 group, TGF-β1+IGF-1+ BMP-2 group, and blank control group. At 21 days of induction, cels were stained with alcian blue and alizarin red; RT-PCR was employed to detect colagen I mRNA expression. RESULTS AND CONCLUSION:Alcian blue staining showed metachromasia in the cytoplasm and mesenchyma, and proteoglycan was expressed green; alizarin red staining showed no orange calcium nodules. The bone marrow mesenchymal stem cels were preliminarily deduced to diferentiate into chondrocytes, but could not express the cel phenotypes of bone cels. In the blank control group, the expression of colagen I mRNA was negative. Compared with the TGF-β1 group, the mRNA expression of colagen I was lower in the BMP-2+IGF-1 group, but higher in the TGF-β1+BMP-2 group and TGF-β1+IGF-1+BMP-2 group (P < 0.05). Moreover, the expression of colagen I mRNA was highest in the TGF-β1+IGF-1+BMP-2 group (P < 0.05). These findings indicate that TGF-β1 alone is able to induce the chondrogenic diferentiation of bone marrow mesenchymal stem cels, and the combination of TGF-β1, IGF-1 and BMP-2 can play the biggest role to induce the chondrogenic diferentiation of bone marrow mesenchymal stem cels.

ABSTRACT

OBJECTIVETo investigate the survival and development conditions of community-based organizations (CBOs) for HIV/AIDS prevention and control among men who have sex with men (MSM) in Chinese cities including Shanghai, Hangzhou, Chongqing.

METHODSThis study employed both qualitative (focus groups) and quantitative (questionnaire survey) methods to obtain information from 15 MSM CBOs in three Chinese cities.

RESULTSThe mean work time of the 15 CBOs for HIV/AIDS prevention and control among MSM was 6.7 years (2.1-11.3 years), and the majority of their funds was from international cooperation projects (80 447 000 RMB, 73.0%) from 2006 to 2013. The survival cost of MSM CBOs apart from expenditure of activities was 2 240-435 360 RMB per year. As it was shown in the graph, the survival and development of MSM CBOs was closely related to the development of international cooperation projects. There was a few small size MSM CBOs taking part in the prevention and control of HIV/AIDS and their work content was limited before 2006. From 2006 to 2008, some international cooperation projects were launched in China, such as the China Global Fund AIDS project and the China-Gates Foundation HIV Prevention Cooperation program. As a result, the number of MSM CBOs was increased sharply, and both the scale and 2012, the performance of these programs further promote the establishment of new MSM CBOs and the development of all MSM CBOs with regard to the work places, full-time staffs, work contents, work patterns and the specific targeted population. After 2012, most international cooperation programs were completed and the local department of disease prevention and control continued to cooperate with MSM CBOs. However, the degree of support funds from the local department was different among different regions. Where the funds were below the half of program funds, the development of MSM CBOs ceased and work slowed down. Besides, there were still some constraints for the survival and development of MSM CBOs, such as insufficient funds, no legitimate identity, the outflow of talents and the unsustainable development.

CONCLUSIONThe survival and development of MSM CBOs was closely related to the development of international cooperation projects in China. Some departments of disease prevention and control took over the cooperation with MSM CBOs when the international cooperation projects were completed. Given the survival cost of MSM CBOs and the constraints of MSM CBOs development, it needs further investigation on how to ensure the local departments of disease prevention and control to take over the cooperation with MSM CBOs and how to cooperate with MSM CBOs.

ABSTRACT

OBJECTIVETo explore the features of various mediastinal lymphadenopathies using computed tomography perfusion (CTP).

METHODSCTP parameters (CTPs) of the selected mediastinal nodes from 59 patients with pathology-proven malignant lymph nodes and of those from 29 patients with clinically diagnosed or pathology-proven inflammatory lymphadenopathies were collected. Patients were divided into subgroups by etiology and phase of primary disease, including different pathological malignant nodes and diverse inflammatory nodes. CTPs were defined as blood flow (BF), blood volume (BV), mean transit time (MTT), permeability (PMB), and time to peak (TTP). Differences of CTPs were compared between malignant and benign nodes, and among subgroups, respectively.

RESULTSIn the mediastinum, no significant differences of CTPs were found between malignant and benign groups (all P>0.05), the same for subgroups of malignant nodes (all P>0.05). Acute lymphadenitis had higher BF and BV than chronic inflammatory, lymphoid tuberculosis, sarcoidosis and malignant nodes. The BF of malignant nodes was markedly slower than that of acute lymphadenitis (P=0.01), but faster than chronic inflammatory nodes (P=0.04) and sarcoidosis (P=0.03), with no significant difference compared with lymphoid tuberculosis. Pneumonia-complicated lymphoid tuberculosis showed the longest MTT while sarcoidosis displayed the shortest MTT, and inflammatory nodes, lymphoid tuberculosis without complicated pneumonia and malignant nodes had moderate MTT.

CONCLUSIONCTPs show promising potential in distinguishing various lymphadenopathies in the mediastinum, but more studies are needed to improve their specificity.

ABSTRACT

OBJECTIVE@#To evaluate therapeutic eff ect of siRNA-HDAC5 on non-obese diabetic (NOD) mice by using small interference RNA (siRNA) technique to knock down the expression of HDAC5 in spleen CD4+ T cells.@*METHODS@#NOD mice, 12-weeks old, were randomly divided into 3 groups and were given normal saline, siRNA-Control or siRNA-HDAC5 through caudal vein injection. The spleens and other samples were collected at the 18th, 24th or 30th week. The blood glucose was tested by blood glucose meter. The urinary albumin and serum levels of IL-1, IL-6, IL-18, and TNF-α were detected by ELISA. The mRNA levels of CD11a, CCR5, and CX3CR1 in spleen CD4+ T cells were measured by quantitative Real-time PCR. The HDAC5 protein level in spleen CD4+ T cell was detected by Western blot.@*RESULTS@#Compared with the control group, the siRNA-HDAC5 group showed a significant decrease in blood glucose, urine albumin excretion rate, serum cytokine and the mRNA levels of CD11a, CCR5, and CX3CR1, consist with the decrease in protein level of HDAC5.@*CONCLUSION@#Inhibition of HDAC5 expression in NOD mice could effectively alleviate the onset and development of kidney damage caused by diabetes.
